Description

The ZRANB3 (zinc finger RANBP2-type containing 3) is a protein-coding gene located on chromosome 2.

ZRANB3 is a DNA annealing helicase and endonuclease that plays a critical role in maintaining genome stability during DNA replication. It is recruited to stalled or collapsed replication forks by polyubiquitinated PCNA. ZRANB3 acts as a structure-specific endonuclease, cleaving the replication fork D-loop intermediate to create an accessible 3‘-OH group on the leading strand template. This allows DNA polymerase to continue replication. Additionally, ZRANB3 catalyzes fork regression through its annealing helicase activity, preventing replication fork disintegration and double-strand breaks. These activities help restart stalled replication forks and limit inappropriate recombination during template switching events.
