Description
The WWP1 (WW domain containing E3 ubiquitin protein ligase 1) is a protein-coding gene located on chromosome 8.
The WWP1 gene produces a protein found in various tissues. This protein has E3 ubiquitin ligase activity, which plays a role in targeting other proteins for breakdown within cells. WWP1 binds to the PTEN enzyme, a tumor suppressor that regulates cell division. However, this binding disrupts PTEN's ability to dimerize, impairing its activity and potentially contributing to uncontrolled cell growth.
WWP1 acts as an E3 ubiquitin ligase, accepting ubiquitin from an E2 enzyme and transferring it to target proteins. This process can lead to the degradation of various proteins, including ERBB4 isoforms (JM-A CYT-1 and JM-B CYT-1), KLF2, KLF5, and TP63. Interestingly, WWP1 can ubiquitinate RNF11 without triggering its degradation. WWP1 also promotes the degradation of TGFBR1, with the process being enhanced by SMAD7. Additionally, it ubiquitinates SMAD6 and SMAD7. Furthermore, WWP1 ubiquitinates and promotes the degradation of SMAD2 in response to TGF-beta signaling, requiring interaction with TGIF.
