WD repeat-containing protein 46 is a protein that in humans is encoded by the WDR46 gene.
WDR46 acts as a scaffold within the nucleolus, a structure within the nucleus. It plays a crucial role in directing DDX21 and NCL, two other proteins, to a specific region of the nucleolus called the granular compartment. WDR46 is also a component of the small subunit (SSU) processome, a complex responsible for assembling the small ribosomal subunit, a key component of the protein synthesis machinery. This process involves a coordinated effort by numerous proteins, an RNA chaperone, and ribosomal proteins that bind to the nascent pre-ribosomal RNA (pre-rRNA). Their combined action results in the precise folding, modification, rearrangement, and cleavage of the pre-rRNA, along with the targeted degradation of any unwanted pre-ribosomal RNA by the RNA exosome.
