Description

The VNN2 (vanin 2) is a protein-coding gene located on chromosome 6.

VNN2 is a protein encoded by the VNN2 gene in humans. It belongs to the Vanin family of proteins, which share extensive sequence similarity with each other and with biotinidase. The family includes secreted and membrane-associated proteins, some of which are involved in hematopoietic cell trafficking. While no biotinidase activity has been demonstrated for vanin proteins, they possess pantetheinase activity, potentially playing a role in oxidative-stress response. VNN2 is a GPI-anchored cell surface molecule that contributes to the transendothelial migration of neutrophils. The VNN2 gene is located near two other vanin genes on chromosome 6q23-q24, with the same transcriptional orientation. Two transcript variants encoding different isoforms have been described for this gene.

VNN2 is an amidohydrolase that specifically hydrolyzes one of the carboamide linkages in D-pantetheine, recycling pantothenic acid (vitamin B5) and releasing cysteamine. It plays a role in the thymus homing of bone marrow cells and may regulate beta-2 integrin-mediated cell adhesion, migration, and motility of neutrophils.

VNN2 is also known as FOAP-4, GPI-80.
