Description
The USP5 (ubiquitin specific peptidase 5) is a protein-coding gene located on chromosome 12.
Ubiquitin specific peptidase 5 is an enzyme that in humans is encoded by the USP5 gene.
USP5 is an enzyme that cleaves both linear and branched multiubiquitin chains, with a strong preference for branched chains. It plays a crucial role in breaking down unanchored 'Lys-48'-linked polyubiquitin chains, a process important for regulating protein degradation. USP5 also binds to linear and 'Lys-63'-linked polyubiquitin chains, though with a lower affinity. When USP5 activity is reduced, it leads to an accumulation of p53/TP53 protein and increased p53/TP53 transcriptional activity. This occurs because the unanchored polyubiquitin that accumulates can compete with ubiquitinated p53/TP53 for proteasomal recognition, but not with MDM2.
