Description

The TTLL7 (tubulin tyrosine ligase like 7) is a protein-coding gene located on chromosome 1.

TTLL7 is a polyglutamylase that adds polyglutamate side chains to tubulin. These side chains vary in length and are attached to the gamma-carboxyl group of specific glutamate residues within the C-terminal tail of tubulin. TTLL7 initiates and elongates the polyglutamylation reaction in an ATP-dependent manner, preferentially modifying the beta-tubulin tail over the alpha-tail. It competes with TTLL3, a monoglycylase, for the same modification site on beta-tubulin, resulting in an inverse relationship between glycylation and glutamylation. TTLL7 is crucial for neurite growth and contributes to the significant increase in tubulin polyglutamylation observed during the postnatal development of neurons.
