TRIM69


Description

The TRIM69 (tripartite motif containing 69) is a protein-coding gene located on chromosome 15.

TRIM69 is a protein encoded by the TRIM69 gene in humans. It belongs to the RING-B-box-coiled-coil (RBCC) family and features an N-terminal RING finger motif, a PRY domain, and a C-terminal SPRY domain. The mouse ortholog of this gene is specifically expressed in germ cells during spermatogenesis and induces apoptosis when overexpressed. Different isoforms of the protein are produced due to alternative splicing.

TRIM69 is an E3 ubiquitin ligase that plays a crucial role in antiviral immunity by limiting viral infection. It directly targets viral proteins, such as dengue virus NS3, through ubiquitination. TRIM69 also acts as a key mediator of type I interferon-induced microtubule stabilization, independently of its E3 ligase activity. Further, TRIM69 is involved in cataract formation in association with TP53. It inhibits UVB-induced cell apoptosis and ROS production by inducing TP53 ubiquitination. Additionally, TRIM69 regulates centrosome dynamics and mitotic progression by ubiquitinating STK3/MST2, leading to its redistribution to the perinuclear cytoskeleton and subsequent phosphorylation by PLK1.
