The TRAM2 (translocation associated membrane protein 2) is a protein-coding gene located on chromosome 6.
TRAM2 is a component of the translocon, a specialized channel at the endoplasmic reticulum (ER) membrane. The translocon helps control the post-translational processing of newly made secretory and membrane proteins.
TRAM2 plays a crucial role in collagen type I synthesis. It may connect the activity of the ER calcium pump (SERCA2B) with the translocon, leading to a higher local calcium concentration at the collagen synthesis site. This elevated calcium is likely essential for the function of chaperones involved in collagen folding. Additionally, TRAM2 ensures the correct insertion of the first transmembrane helix N-terminus of TM4SF20 into the ER lumen. It has been suggested that TRAM2 acts as a ceramide sensor for regulated alternative translocation (RAT).
