Description

The TRAF5 (TNF receptor associated factor 5) is a protein-coding gene located on chromosome 1.

TRAF5 is a protein encoded by the TRAF5 gene in humans. It belongs to the tumor necrosis factor receptor-associated factor (TRAF) protein family and contains several domains, including a MATH domain, a RING-type zinc finger, and two TRAF-type zinc fingers. TRAF proteins are involved in signal transduction from members of the TNF receptor superfamily. TRAF5 is a component of a protein complex that binds to the cytoplasmic domains of tumor necrosis factor (TNF) receptors, mediating TNF-induced activation. Different versions of the protein are produced through alternative splicing.

TRAF5 acts as an adapter protein and signal transducer, connecting members of the tumor necrosis factor receptor family to various signaling pathways. This connection is achieved through its association with the receptor cytoplasmic domain and kinases. TRAF5 facilitates the activation of NF-kappa-B and likely JNK, and appears to play a role in apoptosis. It also contributes to the activation of NF-kappa-B by EIF2AK2/PKR.
