TOB1


Description

The TOB1 (transducer of ERBB2, 1) is a protein-coding gene located on chromosome 17.

TOB1 is a human gene that encodes the Tob1 protein, a member of the tob/btg1 family of anti-proliferative proteins. Tob1 regulates cell growth and inhibits T cell proliferation, cytokine and cyclin transcription. It interacts with and enhances the DNA binding activity of SMAD2 and SMAD4, which in turn inhibits interleukin 2 transcription in T cells. Tob1 is phosphorylated by ribosomal S6 kinase (RSK) and interacts with ERBB2, potentially interfering with growth suppression. Tob1 interacts with CNOT7, MAPK1, MARCKS, Mitogen-activated protein kinase 9, and RPS6KA1.

TOB1 acts as an anti-proliferative protein by associating with deadenylase subunits of the CCR4-NOT complex. It mediates CPEB3-accelerated mRNA deadenylation by binding to CPEB3 and recruiting CNOT7, leading to the degradation of target mRNAs.
