Description

The TNFSF9 (TNF superfamily member 9) is a protein-coding gene located on chromosome 19.

Tumor necrosis factor ligand superfamily member 9, also known as 4-1BB ligand or 4-1BBL or CD137L, is a protein encoded by the TNFSF9 gene. It is a type 2 transmembrane glycoprotein receptor found on antigen presenting cells (APCs) and binds to 4-1BB (also known as CD137). The 4-1BB/4-1BBL complex belongs to the TNFR:TNF superfamily, expressed on activated T lymphocytes. The complex consists of three monomeric 4-1BBs bound to a trimeric 4-1BBL. Each 4-1BB monomer binds to two 4-1BBLs via cysteine-rich domains (CRDs). The interaction with the second 4-1BBL is crucial for stabilization. Studies on Ag104A sarcoma and P815 mastocytoma have shown that anti-4-1BB antibodies have potent anti-tumor effects, inhibiting tumor growth by increasing CTL activity. Further studies have verified and legitimized the effect of 4-1BB signaling on tumor growth inhibition.
