Description

The TNFSF4 (TNF superfamily member 4) is a protein-coding gene located on chromosome 1.

OX40L (also known as TNFSF4, CD252, or Glycoprotein Gp34) is the ligand for OX40 (TNFRSF4). It is expressed on antigen-presenting cells like DC2s, macrophages, and activated B lymphocytes. OX40 is found on activated T lymphocytes (mainly CD4+ T cells), NK cells, NKT cells, and neutrophils. When OX40 and OX40L bind, it signals T cell survival and memory T cell development. This interaction also promotes Th2 immune responses, even in low IL-4 environments. OX40L is also present on non-immune cells like endothelial and smooth muscle cells. Its expression is triggered by pro-inflammatory mediators such as TNF-α, IFN-γ, and PGE2. Several single-nucleotide polymorphisms (SNPs) in the OX40L gene have been associated with systemic lupus erythematosus.

TNFSF4 is also known as CD134L, CD252, GP34, OX-40L, OX4OL, TNLG2B, TXGP1.
