The TNFRSF12A (TNF receptor superfamily member 12A) is a protein-coding gene located on chromosome 16.
TNFRSF12A, or Tumor Necrosis Factor Receptor Superfamily Member 12A, is a protein encoded by the TNFRSF12A gene in humans. It is also known as Fibroblast Growth Factor-Inducible Immediate-Early Response Protein 14 (FN14) and is a unique member of the Tumor Necrosis Factor Receptor superfamily. TNFRSF12A is the smallest member of this family and its gene expression is tightly regulated. It is the sole receptor for TWEAK, its ligand. TNFRSF12A is found in various human tissues, including the heart, placenta, lung, skeletal muscle, kidney, and pancreas. It is involved in a number of biological processes, such as inflammation, angiogenesis, cell proliferation, and cell death. In humans, the TNFRSF12A gene is located on chromosome 16, while in mice, it is found on chromosome 17. The TNFRSF12A receptors in mice and humans share a 93% similarity.
TNFRSF12A binds to its ligand, TNFSF12/TWEAK, and triggers a variety of cellular responses. While it can induce apoptosis in some cell types, it is primarily known for promoting angiogenesis (new blood vessel formation) and endothelial cell proliferation. Additionally, it may regulate how cells adhere to extracellular matrix proteins.
