Description

The TNFAIP1 (TNF alpha induced protein 1) is a protein-coding gene located on chromosome 17.

TNFAIP1 is a protein encoded by the TNFAIP1 gene in humans. Its expression can be induced by tumor necrosis factor alpha (TNF) in umbilical vein endothelial cells. Studies on a similar gene in mice suggest that its expression is developmentally regulated in a tissue-specific manner.

TNFAIP1 acts as a substrate-specific adapter within the BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex, playing a crucial role in regulating cytoskeleton structure. This complex specifically targets RHOA for ubiquitination, leading to its degradation via the proteasome. This process ultimately regulates the actin cytoskeleton and cell migration. TNFAIP1's interaction with RHOB may also be involved in regulating apoptosis. Furthermore, TNFAIP1 may enhance the activity of DNA polymerase delta in a PCNA-dependent manner.
