Description

The TMEM68 (transmembrane protein 68) is a protein-coding gene located on chromosome 8.

TMEM68, also known as DIESL, is a key enzyme in the alternative triglyceride biosynthesis pathway. It catalyzes the formation of triacylglycerol (TAG) from diacylglycerol (DAG) and membrane phospholipids, particularly phosphatidylcholine (PC) and its ether-linked form (ePC). This process alters the composition of cellular membranes. The alternative pathway is likely essential for providing energy during rapid growth when fuel sources are limited. TMEM68 also contributes to maintaining mitochondrial function during periods of lipid starvation. In addition to its primary role in TAG synthesis, TMEM68 can utilize acyl-CoA as a donor, exhibiting both acyl-CoA:monoacylglycerol acyltransferase (MGAT) and acyl-CoA:diacylglycerol acyltransferase (DGAT) activity.

TMEM68 is also known as DIESL, MGAT/DGAT.
