Description

The TMED2 (transmembrane p24 trafficking protein 2) is a protein-coding gene located on chromosome 12.

Transmembrane emp24 domain-containing protein 2 is a protein that in humans is encoded by the TMED2 gene.

TMED2 is involved in vesicular protein trafficking, primarily in the early secretory pathway but also in post-Golgi membranes. It acts as a cargo receptor on the lumenal side for incorporating secretory cargo molecules into transport vesicles and contributes to vesicle coat formation on the cytoplasmic side. In COPII vesicle-mediated anterograde transport, TMED2 participates in the transport of GPI-anchored proteins, potentially acting together with TMED10 as their cargo receptor. This function specifically involves SEC24C and SEC24D of the COPII vesicle coat and lipid raft-like microdomains of the ER. TMED2 recognizes GPI anchors structurally remodeled in the ER by PGAP1 and MPPE1. During COPI vesicle-mediated retrograde transport, TMED2 inhibits the GTPase-activating activity of ARFGAP1 towards ARF1, preventing premature uncoating and allowing cargo selection. It also plays a role in trafficking G protein-coupled receptors (GPCRs). TMED2 regulates the exocytic trafficking of F2RL1, OPRM1, and P2RY4 from the Golgi to the plasma membrane, contributing to receptor resensitization. Furthermore, it facilitates CASR maturation and stabilization in the early secretory pathway, enhancing CASR plasma membrane targeting. TMED2 is believed to participate in organizing intracellular membranes, such as maintaining the Golgi apparatus. It may also play a role in the biosynthesis of secreted cargo, such as eventual processing.
