TMED1


Description

The TMED1 (transmembrane p24 trafficking protein 1) is a protein-coding gene located on chromosome 19.

TMED1, also known as transmembrane emp24 domain-containing protein 1, is a member of the p24 protein family. This family comprises transmembrane proteins that are crucial components of COPI and COPII-coated vesicles, involved in protein trafficking between the endoplasmic reticulum and Golgi apparatus. While some p24 proteins are essential for survival, TMED1, specifically, has been found to be involved in the biogenesis of COPI and COPII-coated vesicles. It contributes to transporting membrane-bound proteins through the secretory system and participates in forming the structure of the endoplasmic reticulum and Golgi. TMED1 is widely expressed across various tissues.

TMED1 plays a role in vesicular protein trafficking, particularly in the early secretory pathway. It may act as a cargo receptor at the lumenal side, facilitating the incorporation of secretory cargo molecules into transport vesicles. Additionally, it might be involved in vesicle coat formation at the cytoplasmic side. TMED1 positively contributes to IL-33-mediated IL-8 and IL-6 production by interacting with the interleukin-33 receptor IL1RL1 (PubMed:23319592). Furthermore, TMED1 participates in the modulation of innate immune signaling through the cGAS-STING pathway by interacting with RNF26 (PubMed:32614325).
