Description

The TLN2 (talin 2) is a protein-coding gene located on chromosome 15.

TLN2 is a human protein encoded by the TLN2 gene. It belongs to the talin protein family, closely related to talin-1, a cytoskeletal protein vital for actin filament assembly. TLN2 is predominantly expressed in cardiac muscle, where it forms connections between the extracellular matrix and the actin cytoskeleton at costamere structures, facilitating force transduction.

TLN2 is a large protein, similar in size to talin-1, with a length of 2542 amino acids and a molecular weight of 271.4 kDa. While the protein size is comparable, the TLN2 gene is significantly larger (200 kb) than talin-1 (30 kb) due to differences in intron sizes. TLN2 mRNA is found in various tissues, including cardiac muscle, embryonic stem cells, brain, lung, skeletal muscle, kidney, and testis, with the highest expression in cardiac muscle. The TLN2 gene undergoes complex alternative splicing, generating multiple mRNA transcripts and protein isoforms. A specific promoter associated with a CpG island is responsible for the majority of TLN2 expression in adult tissues. This promoter is located over 200 kb upstream of the first coding exon and is separated from it by a region of alternatively spliced noncoding exons.
