The THBS4 (thrombospondin 4) is a protein-coding gene located on chromosome 5.

Thrombospondin-4 is a protein encoded by the THBS4 gene in humans. It belongs to the thrombospondin protein family, which are adhesive glycoproteins that mediate cell-to-cell and cell-to-matrix interactions. THBS4 forms a pentamer and can bind to heparin and calcium. It may be involved in local signaling in the developing and adult nervous system, in bone formation and fracture healing, and in osteoarthritis.

Thrombospondin-4 (THBS4) is an adhesive glycoprotein that facilitates interactions between cells and the extracellular matrix (ECM). It plays a role in various cellular processes including proliferation, migration, adhesion, and attachment. THBS4 is involved in inflammatory responses to central nervous system (CNS) injuries, regulating vascular inflammation, and adapting the heart to pressure overload. THBS4 binds to structural ECM proteins and modulates the ECM in response to tissue damage, contributing to cardioprotective and adaptive ECM remodeling. It is also involved in the endoplasmic reticulum (ER) stress response by interacting with activating transcription factor 6 alpha (ATF6), a protein that triggers adaptive ER stress responses and protects the myocardium from pressure overload. THBS4 may contribute to spinal presynaptic hypersensitivity and neuropathic pain states after peripheral nerve injury. Additionally, it may regulate protective astrogenesis from the subventricular zone (SVZ) niche after injury in a NOTCH1-dependent manner.
