Description

The TBC1D5 (TBC1 domain family member 5) is a protein-coding gene located on chromosome 3.

TBC1D5 is predicted to function as a GTPase-activating protein (GAP) for members of the Rab family, including RAB7A. It is capable of removing both RAB7A and the retromer CSC subcomplex from the endosomal membrane and relocating them to the cytosol. This retromer displacement appears to rely on TBC1D5's catalytic activity. TBC1D5 is also required for the retrograde transport of cargo proteins from endosomes to the trans-Golgi network (TGN), a process that likely depends on its catalytic activity. TBC1D5 plays a role in regulating autophagy, potentially serving as a molecular switch between endosomal and autophagosomal transport. This protein participates in the reprogramming of vesicle trafficking upon autophagy induction. It is involved in the trafficking of ATG9A when autophagy is activated and might regulate the recruitment of ATG9A-AP2-containing vesicles to autophagic membranes.
