Description

The TAB3 (TGF-beta activated kinase 1 (MAP3K7) binding protein 3) is a protein-coding gene located on chromosome X.

TAB3 is an adapter protein that plays a critical role in activating the JNK and NF-kappa-B signaling pathways. It achieves this by specifically recognizing 'Lys-63'-linked polyubiquitin chains through its RanBP2-type zinc finger (NZF) domain. This interaction serves as a bridge, linking MAP3K7/TAK1 and TRAF6 to these ubiquitin chains, ultimately facilitating the formation of a large signaling complex. This complex promotes autophosphorylation of MAP3K7/TAK1, which in turn activates the I-kappa-B-kinase (IKK) core complex. This intricate interplay of interactions is crucial for the proper activation of these vital signaling pathways.

TAB3 is also known as MAP3K7IP3, NAP1.
