STEAP3
Description
The STEAP3 (STEAP3 metalloreductase) is a protein-coding gene located on chromosome 2.
STEAP3 is a metalloreductase enzyme encoded by the STEAP3 gene in humans. It converts insoluble ferric iron (Fe3+) to soluble ferrous iron (Fe2+). STEAP3 and other STEAP proteins, except STEAP1, are predicted to have a Di-nucleotide binding domain (Rossmann Fold), confirmed by X-ray crystallography for STEAP3 and STEAP4. STEAP3 interacts with BNIP3L and PKMYT1.
STEAP3 is an integral membrane protein that functions as a NADPH-dependent ferric-chelate reductase, using NADPH from one side of the membrane to reduce a Fe(3+) chelate that is bound on the other side of the membrane. It mediates sequential transmembrane electron transfer from NADPH to FAD and onto heme, and finally to the Fe(3+) chelate. STEAP3 can also reduce Cu(2+) to Cu(1+). It mediates efficient transferrin-dependent iron uptake in erythroid cells. STEAP3 may play a role downstream of p53/TP53 to interface apoptosis and cell cycle progression. It is indirectly involved in exosome secretion by facilitating the secretion of proteins such as TCTP.
