Description

The SPTLC3 (serine palmitoyltransferase long chain base subunit 3) is a protein-coding gene located on chromosome 20.

SPTLC3 is a subunit of the serine palmitoyltransferase (SPT) enzyme complex. SPT catalyzes the initial and rate-limiting step in sphingolipid biosynthesis by combining L-serine with activated acyl-CoA (most commonly palmitoyl-CoA) to produce long-chain bases. The composition of the SPT complex determines substrate preference. The SPTLC1-SPTLC2-SPTSSA complex favors C16-CoA, while the SPTLC1-SPTLC3-SPTSSA isozyme uses both C14-CoA and C16-CoA, with a slight preference for C14-CoA. The SPTLC1-SPTLC2-SPTSSB complex shows a strong preference for C18-CoA, while the SPTLC1-SPTLC3-SPTSSB isozyme uses a broader range of acyl-CoAs without apparent preference.
