Description

The SMURF2 (SMAD specific E3 ubiquitin protein ligase 2) is a protein-coding gene located on chromosome 17.

SMURF2 is an enzyme that is encoded by the SMURF2 gene located on chromosome 17. It is involved in regulating TGF-beta signaling by targeting SMAD proteins for ubiquitination and degradation.

SMURF2 is an E3 ubiquitin-protein ligase that accepts ubiquitin from an E2 ubiquitin-conjugating enzyme and transfers it to target substrates. It interacts with SMAD7 to trigger SMAD7-mediated degradation of the TGF-beta receptor, downregulating TGF-beta signaling. This interaction also activates SMAD7's autocatalytic degradation, which is prevented by AIMP1. SMURF2 forms a complex with phosphorylated SMAD1, SMAD2, and SMAD3, targeting SMAD1 and SMAD2 for ubiquitination and degradation. SMAD2 can recruit substrates, such as SNON, for ubiquitination. SMURF2 negatively regulates TGFB1-induced epithelial-mesenchymal transition and myofibroblast differentiation.
