Description

The SLC16A3 (solute carrier family 16 member 3) is a protein-coding gene located on chromosome 17.

Monocarboxylate transporter 4 (MCT4), also known as solute carrier family 16 member 3, is a protein encoded by the SLC16A3 gene. It is widely expressed, particularly in glycolytic tissues like white skeletal muscle fibers, astrocytes, white blood cells, chondrocytes, and some mammalian cell lines. This suggests that MCT4 is well-suited for exporting lactate produced by glycolysis. MCT4 has a lower affinity for most substrates and inhibitors compared to MCT1, with Km and Ki values about 5-10 times higher. Its high Km for pyruvate is significant because it prevents loss of pyruvate from the cell, which would hinder the removal of NADH generated by glycolysis through the conversion of pyruvate to lactate.
