SKAP1
Description
The SKAP1 (src kinase associated phosphoprotein 1) is a protein-coding gene located on chromosome 17.
SKAP1 (Src kinase-associated phosphoprotein 1) is an adapter protein involved in T-cell signaling. It contains a PH domain and a C-terminal SH3 domain, which allow it to interact with other proteins. SKAP1 plays a crucial role in inside-out signaling, coupling T-cell antigen receptor stimulation to the activation of integrins. It is essential for optimal conjugation between T-cells and antigen-presenting cells by promoting the clustering of integrin ITGAL on the surface of T-cells. SKAP1 also regulates LFA-1 adhesion and binds to the exchange factor RasGRP1, regulating ERK activation in T-cells.
SKAP1 positively regulates T-cell receptor signaling by enhancing the MAP kinase pathway. It is essential for optimal conjugation between T-cells and antigen-presenting cells by promoting the clustering of integrin ITGAL on the surface of T-cells. SKAP1 may also be involved in high affinity immunoglobulin epsilon receptor signaling in mast cells.
