SHARPIN


Description

The SHARPIN (SHANK associated RH domain interactor) is a protein-coding gene located on chromosome 8.

The SHARPIN gene encodes a protein that is a component of the LUBAC complex (linear ubiquitin chain assembly complex). This complex is crucial in the regulation of inflammation and cell death by attaching linear polyubiquitin chains to target proteins. It plays a key role in activating NF-kappa-B, a critical transcription factor in immune responses. LUBAC also contributes to the stability of the TNF-R1 signaling complex, which is involved in the cell's response to the tumor necrosis factor. Furthermore, LUBAC is involved in innate immunity by tagging bacteria with ubiquitin chains, marking them for destruction by autophagy. Notably, SHARPIN, in conjunction with OTULIN, regulates the canonical Wnt signaling pathway during angiogenesis.

SHARPIN is also known as AIFID, SIPL1.
