RuvB-like 1 (E. coli), also known as RUVBL1 and TIP49, is a human gene. RUVBL1 can form a hexamer. The hexamer can form a dodecamer with RUVBL2 protein. Possesses single-stranded DNA-stimulated ATPase and ATP-dependent DNA helicase (3' to 5') activity; hexamerization is thought to be critical for ATP hydrolysis and adjacent subunits in the ring-like structure contribute to the ATPase activity.

RUVBL1 possesses single-stranded DNA-stimulated ATPase and ATP-dependent DNA helicase (3' to 5') activity. Hexamerization is crucial for ATP hydrolysis, with adjacent subunits in the ring-like structure contributing to the ATPase activity. RUVBL1 is a component of the NuA4 histone acetyltransferase complex, which activates transcription of specific genes by acetylating histones H4 and H2A. This modification alters nucleosome-DNA interactions and promotes interactions with other transcription-regulating proteins. NuA4 is essential for oncogene-mediated growth induction, tumor suppressor-mediated growth arrest, replicative senescence, apoptosis, and DNA repair. RUVBL1 and RUVBL2, along with EP400, contribute to the NuA4 complex's ATPase and helicase activities. NuA4 also plays a direct role in DNA repair. RUVBL1 is a component of a SWR1-like complex that removes histone H2A.Z/H2AZ1 from the nucleosome. It is also a core component of the INO80 chromatin remodeling complex, which exhibits DNA- and nucleosome-activated ATPase activity and catalyzes ATP-dependent nucleosome sliding. RUVBL1 is essential for oncogenic transformation by MYC and modulates transcriptional activation by the LEF1/TCF1-CTNNB1 complex. RUVBL1 is crucial for cell proliferation and may bind plasminogen at the cell surface to enhance plasminogen activation.
