Description

The RPS6KA4 (ribosomal protein S6 kinase A4) is a protein-coding gene located on chromosome 11.

RPS6KA4, the gene encoding ribosomal protein S6 kinase alpha-4, is a member of the RSK (ribosomal S6 kinase) family. This kinase possesses two catalytic domains and phosphorylates various substrates, including CREB1 and c-Fos. It has been shown to interact with MAPK14. RPS6KA4 plays a role in regulating gene expression by phosphorylating transcription factors and histones.

RPS6KA4 is a serine/threonine kinase that plays a crucial role in regulating gene expression in response to mitogenic and stress stimuli. It phosphorylates the transcription factors CREB1 and ATF1, contributing to the activation of genes involved in cell growth and survival. Additionally, RPS6KA4 regulates the activity of the transcription factor RELA, a component of the NF-κB pathway, and contributes to gene activation through histone phosphorylation. This kinase also acts downstream of the Toll-like receptor TLR4 in macrophages, limiting the production of pro-inflammatory cytokines. This function likely involves the induction of transcription of the MAP kinase phosphatase DUSP1 and the anti-inflammatory cytokine interleukin 10 (IL10) through CREB1 and ATF1.
