RNF34
Description
The RNF34 (ring finger protein 34) is a protein-coding gene located on chromosome 12.
RNF34 is an enzyme that encodes a protein with a RINF finger, a motif involved in protein-protein and protein-DNA interactions. This protein interacts with DNAJA3/hTid-1, a DnaJ protein that modulates apoptosis. Overexpression of RNF34 in Hela cells confers resistance to TNF-alpha-induced apoptosis, suggesting an anti-apoptotic function. RNF34 can be cleaved by caspase-3 during apoptosis. Multiple transcript variants encoding distinct isoforms have been reported.
RNF34 is an E3 ubiquitin-protein ligase that regulates several biological processes by promoting the degradation of various target proteins through the ubiquitin-mediated proteasomal pathway. It directly ubiquitinates caspases 8 and 10, leading to their degradation and negative regulation of cell death in the extrinsic apoptotic pathway. RNF34 may also indirectly regulate the tumor necrosis factor-mediated signaling pathway by ubiquitinating and promoting the degradation of RIPK1. Additionally, it negatively regulates p53 through both direct ubiquitination and degradation, and indirectly by degrading SFN. RNF34 is involved in regulating the metabolism of brown fat cells by ubiquitinating and degrading PPARGC1A. Finally, RNF34 may play a role in innate immunity by ubiquitinating NOD1, leading to its degradation.
