Description

The RNF186 (ring finger protein 186) is a protein-coding gene located on chromosome 1.

RNF186 is an E3 ubiquitin protein ligase involved in several cellular processes. It plays a role in apoptosis by activating an apoptotic signaling pathway triggered by endoplasmic reticulum (ER) stress. RNF186 stimulates the expression of unfolded protein response (UPR) proteins, ubiquitinates BNIP1 to regulate its mitochondrial localization, and induces calcium release from the ER, ultimately leading to cell death. It also contributes to intestinal homeostasis and the clearance of enteric pathogens. Upon NOD2 stimulation, RNF186 ubiquitinates ATF6, an ER stress sensor, promoting the UPR. RNF186 participates in basal autophagy by regulating EPHB2 and EPHB3 ubiquitination. Upon EFNB1 stimulation, it ubiquitinates EPHB2 and recruits MAP1LC3B for autophagy induction. Additionally, RNF186 controls nutrient sensing by ubiquitinating Sestrin-2, a cytosolic leucine sensor and mTORC1 inhibitor.
