Description

The RNF11 (ring finger protein 11) is a protein-coding gene located on chromosome 1.

RNF11 is a protein encoded by the RNF11 gene in humans. It contains a RING-H2 finger motif, which is crucial for protein-protein interactions. RNF11 expression is induced by mutant RET proteins associated with multiple endocrine neoplasia (MEN), a condition caused by germline mutations in the RET gene.

RNF11 plays a crucial role in regulating inflammatory signaling pathways by acting as an essential component of a ubiquitin-editing protein complex. This complex, which also includes TNFAIP3, ITCH, and TAX1BP1, ensures that inflammatory signals are transient. Following TNF stimulation, RNF11 promotes the interaction between TNFAIP3 and RIPK1. TNFAIP3 then deubiquitinates 'Lys-63' polyubiquitin chains on RIPK1 and catalyzes the formation of 'Lys-48'-polyubiquitin chains, leading to the proteasomal degradation of RIPK1. This process effectively terminates the activation of NF-kappa-B, which is triggered by TNF or LPS. Additionally, RNF11 recruits STAMBP to the E3 ubiquitin-ligase SMURF2, promoting its ubiquitination and subsequent degradation by the 26S proteasome.
