REV3L, also known as DNA polymerase zeta catalytic subunit (POLZ), is an enzyme encoded by the REV3L gene. It interacts with Rev7 to form Pol ζ, a B family polymerase lacking 3' to 5' exonuclease activity. Pol ζ is a moderate fidelity polymerase, unable to add nucleotides across DNA lesions but capable of extending from primers with terminal mismatches. This makes Pol ζ essential for translesion synthesis (TLS) as it bypasses lesions in concert with other TLS polymerases. Pol ζ's tolerance for mismatches allows cells to survive with mutations, possibly contributing to evolution. REV3L has been shown to interact with MAD2L2.
REV3L is the catalytic subunit of DNA polymerase zeta, an error-prone enzyme involved in translesion DNA synthesis (TLS). It lacks proofreading capabilities due to the absence of a 3'-5' exonuclease activity.
