Description

The RBBP6 (RB binding protein 6, ubiquitin ligase) is a protein-coding gene located on chromosome 16.

Retinoblastoma-binding protein 6 (RBBP6) is a protein encoded by the RBBP6 gene in humans. It plays a crucial role in cell cycle regulation, acting as a binding partner for the retinoblastoma tumor suppressor protein (pRB). RBBP6 binds to underphosphorylated pRB, which is important for suppressing cellular proliferation. The activity of pRB is regulated by cell cycle-dependent phosphorylation. RBBP6 has been shown to interact with Y box binding protein 1 (YBX1). Multiple alternatively spliced transcript variants of the RBBP6 gene have been found, encoding different isoforms.

RBBP6 is an E3 ubiquitin-protein ligase that promotes the ubiquitination of YBX1, leading to its degradation by the proteasome (PubMed:18851979). It may also act as a scaffold protein, facilitating the assembly of the p53/TP53-MDM2 complex. This complex formation increases MDM2-mediated ubiquitination and degradation of p53/TP53, potentially acting as a negative regulator of p53/TP53, influencing both apoptosis and cell growth (By similarity). RBBP6 plays a role in regulating DNA replication and the stability of chromosomal common fragile sites (CFSs) in a ZBTB38- and MCM10-dependent manner. It controls ZBTB38 protein stability and abundance through ubiquitination and proteasomal degradation. In turn, ZBTB38 negatively regulates the expression of MCM10, which is crucial for DNA replication (PubMed:24726359).
