RAD18
Description
The RAD18 (RAD18 E3 ubiquitin protein ligase) is a protein-coding gene located on chromosome 3.
RAD18 is a human enzyme encoded by the RAD18 gene. It is highly similar to the yeast DNA damage repair protein Rad18, which interacts with Rad6, a ubiquitin-conjugating enzyme involved in post-replication repair of damaged DNA. RAD18 also interacts with the human homolog of yeast Rad6 protein through a conserved ring finger motif. Mutation of this motif leads to defective replication of UV-damaged DNA and increased sensitivity to various mutagens.
RAD18 is an E3 ubiquitin-protein ligase that plays a crucial role in postreplication repair of UV-damaged DNA. This repair process involves filling gaps in the newly synthesized DNA strand during replication when the template strand contains damage. RAD18 forms a complex with the E2 ubiquitin conjugating enzyme UBE2B, and together they mono-ubiquitinate PCNA (proliferating cell nuclear antigen) at Lys-164, which is a key step in the repair process. RAD18 also has the ability to bind single-stranded DNA (ssDNA).
