The PPP1R16B (protein phosphatase 1 regulatory subunit 16B) is a protein-coding gene located on chromosome 20.
PPP1R16B is a regulator of protein phosphatase 1 (PP1) that positively regulates pulmonary endothelial cell (EC) barrier function. It is involved in the PI3K/AKT signaling pathway, angiogenesis, and endothelial cell proliferation, likely through controlling ECE1 dephosphorylation, trafficking, and activity. PPP1R16B protects the endothelial barrier from LPS-induced vascular leakage and regulates endothelial cell filopodia extension. It may be a downstream target for TGF-beta1 signaling in endothelial cells and participates in PKA-mediated moesin dephosphorylation, crucial for EC barrier protection against thrombin stimulation. PPP1R16B promotes the interaction of PPP1CA with RPSA/LAMR1, facilitating RPSA/LAMR1 dephosphorylation. It is involved in the dephosphorylation of EEF1A1.
