The PON3 (paraoxonase 3) is a protein-coding gene located on chromosome 7.

Paraoxonase 3, also known as PON3, is a protein encoded by the PON3 gene in humans. It belongs to the paraoxonase family and is located in a cluster on chromosome 7 with the other two family members. PON3 is secreted into the bloodstream and associates with high-density lipoprotein (HDL). It can rapidly hydrolyze lactones and inhibit the oxidation of low-density lipoprotein (LDL), a function believed to slow the initiation and progression of atherosclerosis. Several alternatively spliced variants have been described, encoding different protein isoforms, but only one has been fully characterized.
