POLDIP2
Description
The POLDIP2 (DNA polymerase delta interacting protein 2) is a protein-coding gene located on chromosome 17.
POLDIP2, also known as PDIP38, is a protein encoded by the POLDIP2 gene in humans. It interacts with the p50 subunit of DNA polymerase delta, as well as with proliferating cell nuclear antigen (PCNA). Some transcripts of the POLDIP2 gene overlap with the gene for tumor necrosis factor, alpha-induced protein 1 (TNFAIP1). POLDIP2 is a moonlighting protein, meaning it performs different functions in different cellular locations. In the nucleus, it interacts with PCNA. In the mitochondrion, it interacts with proteins involved in mitochondrial DNA replication and the AAA+ protease CLPXP, where it modulates both the specificity and stability of this proteolytic machine.
POLDIP2 plays a crucial role in DNA damage tolerance by regulating translesion synthesis (TLS), a process that allows DNA replication to continue despite the presence of DNA damage. This involves the repair of damaged DNA templates, such as those containing 8-oxo-guanine (8oxoG) and abasic sites. POLDIP2 is thought to enhance TLS by stimulating the activity of DNA polymerases involved in this process, including PRIMPOL and polymerase delta (POLD1).
