PIAS2


Description

The PIAS2 (protein inhibitor of activated STAT 2) is a protein-coding gene located on chromosome 18.

E3 SUMO-protein ligase PIAS2 is an enzyme that in humans is encoded by the PIAS2 gene.

PIAS2 functions as an E3-type SUMO ligase, facilitating SUMOylation by stabilizing the interaction between the SUMO E2 ligase UBE2I and the target protein. It also acts as a SUMO-tethering factor, bringing SUMO and target proteins together. PIAS2 plays a significant role in regulating gene expression, acting as a transcriptional coregulator in pathways like STAT, p53, and steroid hormone signaling. Its effect on transcription can be either activation or silencing, depending on the specific cellular context and the PIAS2 isoform involved. However, PIAS2 is generally associated with gene silencing. It binds to SUMOylated ELK1, enhancing its transcriptional activity by preventing HDAC2 recruitment, effectively reversing the SUMO-mediated repression of ELK1. PIAS2-beta, but not PIAS2-alpha, promotes MDM2 sumoylation. PIAS2-alpha promotes PARK7 sumoylation. PIAS2-beta promotes NCOA2 sumoylation more effectively than PIAS2-alpha. PIAS2-alpha sumoylates PML at Lys-65 and Lys-160.
