Description

The PHLPP1 (PH domain and leucine rich repeat protein phosphatase 1) is a protein-coding gene located on chromosome 18.

PHLPP1 is a protein phosphatase that regulates the activity of Akt and protein kinase C (PKC) signaling pathways. It specifically removes phosphate groups from the C-terminal hydrophobic motif of AGC kinases, including Akt2 and Akt3 at Ser-473, PRKCB at Ser-660, and PRKCA at Ser-657. PHLPP1's dephosphorylation of Akt promotes apoptosis and tumor suppression. Additionally, it dephosphorylates STK4, leading to its activation and apoptosis. PHLPP1 is also involved in regulating cap-dependent translation by dephosphorylating RPS6KB1. It has been shown to inhibit cancer cell proliferation and may act as a tumor suppressor. PHLPP1 can also dephosphorylate RAF1, inhibiting its kinase activity and potentially acting as a negative regulator of K-Ras signaling. PHLPP1 plays roles in hippocampus-dependent long-term memory formation, circadian control, and the development and function of regulatory T-cells.
