Description

The PHF12 (PHD finger protein 12) is a protein-coding gene located on chromosome 17.

PHD finger protein 12 is a protein that in humans is encoded by the PHF12 gene.

PHF12 acts as a key scaffolding subunit of SIN3 complexes, contributing to complex assembly by interacting with core subunits, stabilizing the complex, and recruiting the H3 histone tail as a substrate. SIN3 complexes are comprised of a SIN3 scaffold subunit, a catalytic core (HDAC1 or HDAC2), and two chromatin targeting modules. The SIN3B complex specifically represses transcription and counteracts the activity of the histone acetyltransferase EP300 by recognizing H3K27ac marks through PHF12 and utilizing the histone deacetylase HDAC2. The SIN3B complex binds downstream of active gene transcription start sites, mitigating histone acetylation and RNA polymerase II progression within transcribed regions to regulate transcription. PHF12 can also independently repress transcription through the recruitment of TLE5 complexes to DNA. Additionally, it may play a role in ribosomal biogenesis.
