Description

The PELI1 (pellino E3 ubiquitin protein ligase 1) is a protein-coding gene located on chromosome 2.

Protein pellino homolog 1 is a protein that in humans is encoded by the PELI1 gene.

PELI1 is an E3 ubiquitin ligase that attaches ubiquitin molecules to other proteins. It plays a role in the TLR and IL-1 signaling pathways by interacting with a complex that includes IRAK kinases and TRAF6. PELI1 mediates 'Lys-63'-linked polyubiquitination of IRAK1, which activates NF-kappa-B. It also mediates 'Lys-48'-linked polyubiquitination of RIPK3, leading to its degradation by the proteasome. PELI1 preferentially recognizes and degrades the 'Thr-182' phosphorylated form of RIPK3, which negatively regulates necroptosis. PELI1 also mediates 'Lys-63'-linked ubiquitination of RIPK1.
