PARP14


Description

The PARP14 (poly(ADP-ribose) polymerase family member 14) is a protein-coding gene located on chromosome 3.

PARP14 is an ADP-ribosyltransferase that specifically attaches a single ADP-ribose molecule (mono-ADP-ribosylation) to glutamate residues on target proteins. It cannot perform poly-ADP-ribosylation, unlike PARP1 and PARP2. PARP14 has been shown to modify STAT1 at Glu-657 and Glu-705, leading to reduced STAT1 phosphorylation and consequently suppression of pro-inflammatory cytokine production in macrophages stimulated by IFNG. However, the role of ADP-ribosylation in inhibiting STAT1 phosphorylation has been debated, with sumoylation of STAT1 at Lys-703 being proposed as an alternative mechanism. PARP14 also modifies STAT6, enhancing STAT6-dependent transcription and promoting STAT6 phosphorylation in macrophages upon IL4 stimulation. Additionally, it mono-ADP-ribosylates PARP9.
