Description

The PANK4 (pantothenate kinase 4 (inactive)) is a protein-coding gene located on chromosome 1.

PANK4 is a human gene that encodes pantothenate kinase 4, an enzyme involved in the biosynthesis of coenzyme A (CoA). Pantothenate kinases catalyze the initial step in CoA production, which is crucial for various metabolic processes. PANK4 is highly expressed in muscle but is found in all tissues. This enzyme is regulated by feedback inhibition from CoA, ensuring proper CoA levels.

PANK4 acts as a phosphatase, specifically targeting 4'-phosphopantetheine and its oxidized forms (sulfonate or S-sulfonate). This activity is believed to prevent damage within the coenzyme A (CoA) pathway. By hydrolyzing excess 4'-phosphopantetheine, PANK4 can limit its oxidation to harmful forms. Additionally, PANK4 hydrolyzes 4'-phosphopantetheine sulfonate or S-sulfonate, preventing their conversion into inactive forms of CoA and acyl carrier protein. It is suggested that PANK4 contributes to regulating CoA levels within cells.
