The ORC3 (origin recognition complex subunit 3) is a protein-coding gene located on chromosome 6.

ORC3 (also known as ORC3L) encodes a protein that is a component of the origin recognition complex (ORC), a six-subunit protein complex crucial for initiating DNA replication in eukaryotic cells. ORC binds to origins of replication and serves as a platform for assembling other initiation factors, including Cdc6 and Mcm proteins. Studies in Drosophila suggest a potential role for ORC3 in neuronal proliferation and olfactory memory. This gene exhibits alternative splicing, resulting in distinct isoforms.

ORC3 is a component of the origin recognition complex (ORC), which binds origins of replication. This binding requires ATP. While the specific DNA sequences that define origins of replication remain unidentified, ORC is essential for assembling the pre-replication complex, a crucial step in initiating DNA replication. ORC3 has been shown to interact with histone H3 and H4 trimethylation marks, including H3K9me3, H3K27me3, and H4K20me3.
