Description
The OAS3 (2'-5'-oligoadenylate synthetase 3) is a protein-coding gene located on chromosome 12.
The OAS3 gene encodes the enzyme 2'-5'-oligoadenylate synthetase 3, which is part of the 2', 5' oligoadenylate synthase family. This enzyme is induced by interferons and catalyzes the synthesis of 2', 5' oligomers of ATP. These oligomers activate latent RNase L, resulting in the degradation of both viral and endogenous RNA. This enzyme family plays a crucial role in inhibiting cellular protein synthesis in response to viral infection.
OAS3 is an interferon-induced, dsRNA-activated antiviral enzyme that plays a crucial role in the innate antiviral response. It synthesizes 2'-5'-oligoadenylates (2-5A) from ATP, which activate ribonuclease L (RNase L), leading to viral and cellular RNA degradation and inhibition of protein synthesis, ultimately terminating viral replication. OAS3 can mediate its antiviral effects through the classical RNase L-dependent pathway or an alternative RNase L-independent pathway. It exhibits antiviral activity against Chikungunya virus (CHIKV), Dengue virus, Sindbis virus (SINV), and Semliki Forest virus (SFV).
