Description

The NUDT4 (nudix hydrolase 4) is a protein-coding gene located on chromosome 12.

NUDT4 (Nucleoside diphosphate-linked moiety X motif 4) encodes Diphosphoinositol polyphosphate phosphohydrolase 2, an enzyme that regulates the turnover of diphosphoinositol polyphosphates. These high-energy diphosphoinositol polyphosphates act as molecular switches, potentially regulating intracellular trafficking. The gene exhibits multiple alternatively spliced transcript variants, with some lacking full-length determination. Two isoforms, DIPP2alpha and DIPP2beta, differ only by an additional amino acid in DIPP2beta due to alternate splicing.

NUDT4 cleaves a beta-phosphate from the diphosphate groups in PP-InsP5 (diphosphoinositol pentakisphosphate), PP-InsP4 and [PP]2-InsP4 (bisdiphosphoinositol tetrakisphosphate), suggesting a role in signal transduction. It also hydrolyzes diadenosine 5',5'''-P1,P6-hexaphosphate (Ap6A) but not diadenosine 5',5'''-P1,P5-pentaphosphate (Ap5A), producing ADP and p4a from Ap6A. Additionally, NUDT4 can hydrolyze 5-phosphoribose 1-diphosphate. It does not appear to be involved in U8 snoRNA decapping activity but binds U8 snoRNA.

NUDT4 is also known as DIPP-2B, DIPP2, DIPP2alpha, DIPP2beta, HDCMB47P, NUDT4B.
