Description

The NPRL3 (NPR3 like, GATOR1 complex subunit) is a protein-coding gene located on chromosome 16.

The NPRL3 gene provides instructions for making a protein that is part of the GATOR1 complex. This complex regulates the mTOR pathway, which controls cell growth, division, survival, and protein synthesis. The GATOR1 complex inhibits the mTOR pathway by blocking the activity of mTORC1. In the brain, the mTOR pathway regulates nerve cell growth, development, and plasticity.

NPRL3, as part of the GATOR1 complex, inhibits the amino acid-sensing branch of the mTORC1 pathway. When amino acids are scarce, GATOR1 activates a GTPase called RagA/RRAGA (or RagB/RRAGB), converting it to an inactive form. This disrupts the mTORC1 pathway, preventing its signaling. However, GATOR1 is negatively regulated by GATOR2 when amino acids are plentiful.
