NLRP2
Description
The NLRP2 (NLR family pyrin domain containing 2) is a protein-coding gene located on chromosome 19.
NLRP2 is a protein encoded by the NLRP2 gene in humans. It is a member of the NLR family, characterized by an N-terminal pyrin domain (PYD) and is involved in the activation of caspase-1 (CASP1). It may also be involved in protein complexes that activate proinflammatory caspases. The NLRP2 gene contains a pyrin effector domain (PYD), a nucleotide-binding and oligomerization domain (NACHT), and leucine-rich repeats (LRR). It interacts with components of the IKK complex and regulates the activities of caspase-1 and NF-κB. The pyrin domain is crucial for suppressing NF-κB activity, and a variant in the NLRP2 gene blocks NF-κB transcriptional activity. NLRP2 contributes to the regulation of immune responses.
NLRP2 inhibits NF-κB activation triggered by TNF and CD40 by preventing the breakdown of NFKBIA, a protein that inhibits NF-κB. When NLRP2 interacts with PYCARD, it activates caspase-1, leading to the release of mature IL-1β, a pro-inflammatory cytokine. NLRP2 may be part of the inflammasome, a protein complex that also contains PYCARD, CARD8, and caspase-1. The inflammasome is involved in activating pro-inflammatory caspases.
