Description

The NAE1 (NEDD8 activating enzyme E1 subunit 1) is a protein-coding gene located on chromosome 16.

NAE1 is the regulatory subunit of the dimeric UBA3-NAE1 E1 enzyme. This enzyme activates NEDD8 by a two-step process: first, it adenylates the C-terminal glycine residue of NEDD8 using ATP. Then, it links this residue to the catalytic cysteine of UBA3, creating a NEDD8-UBA3 thioester and free AMP. Finally, E1 transfers NEDD8 to the catalytic cysteine of UBE2M. NAE1 is essential for cell cycle progression through the S-M checkpoint. Overexpression of NAE1 leads to apoptosis by disrupting the regulation of NEDD8 conjugation. The covalent attachment of NEDD8 to target proteins, known as 'neddylation', plays a crucial role in controlling cell growth, survival, and development.
